Obtaining of stable tobacco breeding lines with resistance factors to different PVY isolates from the wild species Nicotiana africana Merxm.

Abstract

The investigations on the transfer of Potato virus Y (PVY) resistance from the wild species Nicotiana africana Merxm. to cultivated tobacco were conducted in the BC2–BC2F6 generation. Cytological investigations covered counts of mitotic chromosomes, meiotic configurations and pollen viability in the successive hybrid generations. A number of mitotic chromosomes varied in the generations studied. As N. africana chromosomes were eliminated, there was increase in the pollen viability. Stable 48-chromosome lines were found in the BC2F4 generation. In the resistance studies using a very virulent strain YNZ the increase in the percentage of resistant plants with each of the advancing generations BC2, BC2F1 and BC2F2 was observed. As the result of selection for resistance to PVY, carried out in further generations BC2F3 – BC2F6 in the conditions of natural field infection, breeding lines expressing the resistance to or tolerance of the occurring PVY isolates were selected. Plants of the BPA line inoculated using six different isolates representing different strains failed to develop necrotic symptoms, but some of them showed vein clearing, and virus was detected by serological methods. Resistance ratings of the breeding lines appeared to be higher than those of N. tabacum cultivars, as it was seen in the absence of necrotic reaction. However, they were lower than that in the N. africana donor species. Since infection pressure of the isolates producing necrotic symptoms in N. tabacum is currently relatively high, the stable breeding lines that carry N. africana-derived resistance factors to various PVY strains/isolates, are the source of a very valuable germplasm to be used in breeding.
