The new alloplasmic Nicotiana tabacum L. line with Nicotiana wuttkei Clarkson et Symon cytoplasm

Abstract
Cytoplasmic-nuclear male sterility (cms) is an important biological tool, which has been used by plant breeders to produce commercial hybrid cultivars. A new source of cytoplasmic male sterility in tobacco with interspecific origin is reported. The new wild species Nicotiana wuttkei Clarkson et Symon (2n = 32), which was discovered in Australia in 1984, is used as the donor of cytoplasm. The nuclear donor is N.tabacum L. cv. Wiślica (2n = 48). Alloplasmic N.tabacum cv. Wiślica cms wuttkei was developed in a backcrossing program from the amphidiploid N.wuttkei × N.tabacum (2n = 80), developed in a previous study. In the first step, sesquidiploid plants (2n = 64) were produced, at 0% to 40% pollen germination. Sesquidiploid plants were back-crossed with the N.tabacum cv. Wiślica for six successive generations and an alloplasmic line were obtained. The plants of the alloplasmic line were male sterile, while their female fertility was normal. The phenotypic effect of N.wuttkei cytoplasm included ragged corolla, protruding stigma, absence of stamens and modified plant habit. Some growth and development traits of N.tabacum cv. Wiślica cms wuttkei were compared with those of commercially exploited tobacco alloplasmic lines. Plants of cms wuttkei were shorter, had fewer and smaller leaves and flowered later than those of cms tabacum-mutant, cms bigelovii or cms suaveolens.
