Analysis of some traits on phenotypic and molecular level for two- and multi-rowed doubled haploids of spring barley (Hordeum vulgare L.)

Abstract

Variability of some quantitative morphological traits and RAPD markers was studied in spring barley doubled haploid (DH) lines obtained earlier in the Department of Quantitative Genetics (Institute of Plant Genetics in Poznań) with use of the Hordeum bulbosum method. Twenty lines were derived after mutagenic treatment of seed set on F1 hybrids between the two-rowed cultivar Maresi and the six-rowed one Klimek. Another twenty lines produced from the same F1 material without mutagens served as the control. Variation of the morphological traits was estimated basing on field trial data. In the molecular studies eleven PCR-RAPD primers were applied and 87 amplification products were obtained; 87.4% of them were polymorphic. The DH lines obtained with mutagenic treatment showed higher variability of the quantitative traits than the control ones. The same was true, however less distinct, for variability on the molecular level. It indicates, that apart from the variation coming from recombination, an additional variation has been expressed, which may be attributed to the mutation effects.
