Abstract

Two hop progenies from the crosses between hop cv. Wye Target, used as a source of R2 gene determining resistance to hop powdery mildew (Podosphaera macularis), and two susceptible male plants were the objects of the work. The susceptibility of hop genotypes to powdery mildew was assessed using two methods: screening hop cones for disease symptoms in the field under natural infection pressure and artificial inoculation of detached hop leaves in Petri dishes, in controlled conditions. Both assessment methods gave the same results of resistance classification in 88% of individuals. The number of plants to be assessed in the field for cone infection would have been reduced by over 70% using the detached leaves inoculation in the early stage of selection. Random amplified polymorphic DNA (RAPD) markers were used to determine the presence of resistance gene. Until now 40 primers were tested. We report here the identification of two RAPD markers closely linked to a gene for resistance to Podosphera macularis.
