Abstract

Identification of 92 potato cultivars based on the electrophoretic pattern of protease inhibitors isolated from potato tubers has been done. Protease inhibitors are proteins that display composition specific for each potato cultivar and therefore can be used as biochemical markers for potato cultivars. Electrophoretic analysis of the proteins was carried out using a fraction of inhibitors separated from other proteins by affinity chromatography. Affinity columns based on Sepharose matrix were prepared by immobilization of trypsin. For binding of protease inhibitors to immobilized trypsin, for samples of potato juice were poured on the column at neutral pH. After washing out the unbound proteins, the trypsin inhibitors were eluated from the column with buffer pH 1.8. The protein solutions were neutralized and then resolved on polyacrylamide gel. Different electrophoretic bands of inhibitors, depending on the size, charge and molecular properties of particular proteins were obtained. Electrophoretic patterns obtained in the work allow identifying all of the tested potato cultivars. Protease inhibitors isolated from cv. Baszta were used as the internal control for easier identification of electrophoretic bands as well as to portray gel to gel discrepancies.
