A consideration on genetic mapping of QTL responsible for the yellow-seedness in winter rapeseed (Brassica napus L.)

Abstract
The yellow seeds of rapeseed contain less fibre and are characterized by higher fat and protein content than the dark seeds. These features have a positive influence on seed performance in oil pressing and on feeding value of the meal and mill cake. However, the resistance to pathogens and the values of mechanical traits are comparatively lower. Moreover, the substantial change in genotype effects in the decrease in yielding level of the yellow-seeded plants. Genetic characterization of double low (00) yellow-seeded lines obtained at the Department of Genetics and Breeding of Oilseed Crops of the Plant Breeding and Acclimatization Institute can contribute to improving the strategy of breading of yellow-seeded varieties. The use of molecular markers linked to specific traits is a simple way to fast and precise selection of desired genotypes. For mapping and searching for markers linked to the yellow-seedness, two populations were developed from two reciprocal crossings of two yellow-seeded doubled haploid (DH) lines and two DH black-seeded lines. Plants from both populations, each consisted of about one hundred individuals, have already been analyzed phenotypically and mapped genetically. For the latter task, several dozen standard Operon RAPD primers and eleven RAPD primers, which are described in literature as generating markers linked to the colour of seeds, were chosen. The results obtained hitherto strongly suggested that the majority of the primers used generate polymorphic products of DNA amplification. In further studies it is planned to use thirty pairs of AFLP primers, fifteen of which have been described to generate markers linked to the colour of seeds. Moreover, the genome map will be supplemented by the data from analysis done using microstatellites. Such a detailed genetic map of the genome of yellow-seeded lines with markers linked to important qualitative genes will be compared with other genetic maps of oilseed rape.
