Efficiency of the SCAR markers from 4RL chromosome in selection of male fertile genotypes and their relation to some morphological traits in rye (Secale cereale L.)

Abstract

The study aimed at a verification if previously published as well as newly developed SCAR markers located on the 4RL chromosome can be efficient in selecting male fertile genotypes with the Pampa cytoplasm. Research material constituted a F2 population developed after crossing the male sterile inbred line 541 with a single randomly chosen plant from the IRAN IX population of primitive rye. Correlations between male fertility and some morphological traits of plants i.e.: number of spikelets per ear, length of ear and plant height were also examined but no strong dependences were detected. Only 4 out of 14 tested markers mapped on the long arm of 4R chromosome revealed polymorphism between parental genotypes of the studied hybrid. These four SCAR markers formed one linkage group. The constructed genetic map covered the distance of 24cM and all markers belonging to this linkage group showed a statistically significant relation to male fertility of the studied individuals. The most effective tool for selection of male fertile genotypes proved to be the SCP16M58 marker. On average, in the analyzed population, seed set under bags was four times higher on plants with the marker band vs. individuals without the band. This marker showed as well a significant relation to the locus controlling plant height, although the correlation coefficient between male fertility of plants and their height was rather low.
