Application of ISSR markers in analysis of intraspecific similarity of Avena sterilis L.

Abstract
The Institute of Genetics, Breeding and Plant Biotechnology of University of Life Sciences in Lublin conducted a program of Avena sativa L. cultivars crossing with wild species of the genus Avena L. Polish and American cultivars and breeding lines were the maternal forms for crossing. The paternal components were wild hexaploid species: A. sterilis L., A. fatua L. and tetraploids: A. murphyi Ladiz., A. maroccana Gdgr. Analysis of ISSR markers polymorphism was conducted in order to determine genetic diversity of the A. sterilis genotypes used in crossing. The ISSR method is much effective in identification of DNA polymorphism and can be employed with success to evaluate the A. sterilis L. genetic diversity. Genetic similarity of the analyzed genotypes, determined based on the ISSR markers polymorphism, ranged from 0.512 (AVE 2532 vs. AVE 2116) to 0.878 (CN 26025 vs. AVE 531), on the average — 0.715. Such values of genetic similarity indices testify to high diversity of the A. sterilis genotypes designated for crossing.
